What is MT1?

MT1 (Melanotan I, international non-proprietary name afamelanotide) is a synthetic peptide of 13 amino acids. It is derived from endogenous α-MSH and contains two targeted substitutions: norleucine at position 4 and D-phenylalanine at position 7.

These changes make the molecule considerably more resistant to enzymatic degradation than the natural hormone. It was developed at the University of Arizona in the 1980s.

MT1 in research

MT1 acts mainly as an agonist at the melanocortin-1 receptor (MC1R). In cell models with melanocytes it is used to study the regulation of melanin synthesis.

Compared with the cyclic MT2, MT1 is considered more selective for MC1R. This property makes it interesting for comparative studies of melanocortin pharmacology.
